My Possible East Coast Trip

The lynch pin here is the Ark encounter, if you are making a trek out to Kentucky and Ohio it is pretty easy to add in Cincinnati Zoo to the trip, if you are not then it does not make sense to drive out to KY and OH this time around.

If you are not going to KY for the Ark then going east from TN/Western NC to the NC zoo makes the most sense. NC Zoo is huge and done really well, it is one of the top 10 in the USA IMO.

National makes the most sense overall since you are going to be in DC and it is a great zoo that is free and easy to get to.

Columbus is close enough to The Wilds and Dayton Zoo. A little further afield you could head to Akron Zoo, Cleveland Zoo, or the other way down to Cincinnati.
